- If the sample is liquid, use a sterile pipette to inoculate a drop of the sample into the sterile culture tube.
- In cases where sampling is done by a swab, after inoculating the swab on the solid medium in the plate, place the swab into the tube containing the liquid medium and incubate.
Results and Interpretation
If bacteria grow on this medium, turbidity will appear after incubation. After this step, the Gram staining shloud be performed and bacteira should be cultivated in suitable media such as Trypticase Soy Agar, 5% Sheep Blood Agar, Chocolate Agar, EMB Agar, and MacConkey Agar. For the cultivation of anaerobic bacteria, an anaerobic gas pack system should be used.
Note
- If the amount of sodium chloride (6.5%) in this medium is increased, it can be used to differentiate between Enterococci and Streptococci. After 42-48 hours, Enterococci will grow in a liquid medium containing 6.5% salt, and other non-Enterococcal group D Streptococci will not grow in this medium after 48 hours.
- According to ISO 6888-1 standard, by adding rabbit plasma to this medium, it can be used to confirm the presence of coagulase-positive Staphylococci in food and dairy products.
