  • Shake the container well. Dissolve 31.5 grams of the medium in 1 liter of distilled water.
  • Heat until the medium powder is completely dissolved in water.
  • Dispense the prepared medium solution into test tubes containing Durham tubes.
  • Autoclave at 121 degrees Celsius for 15 minutes.
  • The prepared medium is stable for up to one week at room temperature in the dark.

STORAGE:

The medium powder should be kept tightly closed and stored at temperatures below 30 degrees Celsius, and the medium should be stored at a temperature of 2-8 degrees Celsius. The best time to use is before the expiration date printed on the product label.

  • Inoculate the samples into culture medium tubes according to the MPN protocol in the reference standard.
  • Incubate the inoculated tubes for 3 hours at a temperature of 35.5 ± 0.5 °C.
  • Then transfer the tubes to a water bath with a temperature of 44.5 ± 0.2 °C and incubate for an additional 21-23 hours.
  • Note that the water level in the water bath should be higher than the level of the culture medium in the tube.

Results and Interpretation

The gas formation inside the Durham tube indicates the presence of intestinal coliforms in the tested sample. The MPN test can be used to determine the number of intestinal coliforms in the sample.

Note

  • Note that the number of coliforms counted is usually higher than the number of E. coli counted in the sample.
  • The interpretation of results obtained with the A-1 culture medium requires knowledge of the microflora of the sample being examined.

  • Prepared Appearance: Light amber, clear, may have a flocculent precipitate.
  • Cultural Response: The medium was prepared according to label directions. After inoculating with below microorganisms, and incubating at 35 ± 2°C for 3 hours, tubes were transfer to a 44.5°C water bath for 21 ± 2 hours. Cultural characteristics were afterwards.
