  • Shake the con tainer of Bile Esculin Azide Agar well. Dissolve 54.65 grams of the powder medium in one liter of distilled water and mix thoroughly.
  • Heat it gently and boil for one minute to completely dissolve the medium.
  • Autoclave the culture medium at 121 degrees Celsius for 15 minutes.
  • The prepared medium can be stored in a plate at a temperature of 2-8°C for up to 2 weeks.

STORAGE:

Store the powder in the bottle tightly closed and below 30°C and the prepared medium in the tubes at 2-8°C. The best time to use is before the expiration date printed on the product label.

  • In accordance with the type of sample and the relevant standard protocol, inoculate the plate containing the culture medium.
  • Incubate the culture plates at a temperature of 35 ± 2°C for 18-24 hours.

Results and Interpretation

  • If the cultivation is done on a plate, any blackening of the color of the cultivation medium in the plate indicates a positive reaction, and the lack of blackening indicates a negative reaction.
  • If the sample is cultured in the form of a slant, and if half of the slant has turned black after 24-48 hours, the result of the test is positive. If less than half of it has turned black after this period, or if it has not turned black at all, the test result will be negative.
