Brilliant-green Phenol-red Lactose Sucrose Agar

Application: The selective medium for isolating Salmonella, except for S. typhosa and Shigella, from pathological samples, feces, urine, food, etc.

Preparation

  • Weigh 57 grams of the powder and dissolve it in 1 liter of distilled water.
  • Heat it gently for 1 minute until the medium is completely dissolved.
  • Autoclave in 121 degree Celsius for 15 minutes.
  • After autoclave allow it to cool down to a temperature of 50 degrees Celsius and pour in strile petri dishes.

 

STORAGE:

Store dehydrated medium below 30 °C and prepared medium at 2-8 °C. The best time to use is before the expiration date printed on the product label.

Test

  • Inoculate the plate either directly with the sample or with samples taken from a rich culture medium.
  • Tests should be performed with culture media that have fewer inhibitors.
  • Incubate the inoculated plates at a temperature of 35 ± 2 degrees Celsius for 24 hours.

Results and Interpretation

  • Lactose and sucrose fermenting bacteria such as E. coli, Citrobacter, Klebsiella, cause the color of the medium to turn yellow and also form yellow colonies.
  • Bacteria that do not ferment lactose and sucrose such as Salmonella can be identified by the pink colony color on the red medium.
  • The growth of Staphylococcus aureus, Enterococcus faecalis, and Bacillus subtilis can be identified by the yellow color change of the medium and the formation of yellow colonies.
