Adding blood to the base culture medium provides many essential growth factors for fastidious fungi. Since many organisms require initial carbon dioxide for growth after inoculation onto the culture plate, it is better to incubate the inoculated plates in an environment with 10-5% carbon dioxide for 24-48 hours at a temperature of 35±2 degrees Celsius.
To isolate fungi from infected samples, it is better to use another non-selective culture medium alongside this medium and incubate at a temperature of 25-30 degrees Celsius. In addition, to isolate systemic mycotic fungi, two sets of culture media should be incubated, one at a temperature of 25-30 degrees Celsius and the other at a temperature of 35±2 degrees Celsius.
Results and Interpretation
After the incubation and growth of the fungi, the colonies should be examined for color and morphology and biochemical and serological tests should be used for better identification. All cultures should be examined weekly for fungal growth, and growth-negative plates can only be reported negative after 4-6 weeks of incubation.
The BHI Agar medium isolated in a tube is used for cultivating and maintaining pure microbial cultures.
