Application: A highly selective medium for isolating Salmonella bacteria species, excluding Salmonella typhi from meat, food, and other samples.

  • Shake the container well. Dissolve 50 grams of the medium powder in 1 liter of distilled water and mix thoroughly.
  • Boil the medium for 1 minute until completely dissolved.
  • Autoclave the culture medium at 121 degrees Celsius for 15 minutes.

STORAGE:

The medium powder should be kept tightly closed and stored at temperatures below 30 degrees Celsius, and the medium should be stored at a temperature of 2-8 degrees Celsius. The best time to use is before the expiration date printed on the product label.

  • Following the standard principles, inoculate the sample on the culture plate and obtain pure colonies.
  • It is recommended to use semi-selective and non-selective media alongside the mentioned culture medium to increase the chances of obtaining gram-negative bacteria, which are present in small amounts in the sample, and also to identify different populations of organisms present in the sample.
  • Incubate the plates away from light at a temperature of 35±2 for 24-18 hours. If the results are negative after 24 hours, continue the incubation for another 24 hours.

Results and Interpretation

After incubation, the expected results from the growth of organisms are as follows:

  • Salmonella species, excluding Salmonella typhi and Salmonella paratyphi: white to pink colonies surrounded by a red halo.
  • Salmonella typhi and Salmonella paratyphi: weak growth or no growth.
  • Shigella: weak growth or no growth.
  • E. coli, Enterobacter, and Klebsiella: yellow or greenish-yellow colonies surrounded by a greenish-yellow halo.
  • Proteus: weak growth or no growth.
  • Pseudomonas: Pink to reddish colonies.
  • Gram-positive bacteria: weak growth or no growth.
